7-Amino-4-methylcoumarin (AMC substrates)
7-Amino-4-methylcoumarin or AMC based enzyme substrates are colourless non-fluorescent products, which on cleavage by an enzyme produce a detectable, soluble, highly fluorogenic endpoint. Fluorescent free AMC has an excitation wavelength of 390nm and emission 460nm.
AMC substrates are used in amino peptidase assays. When the amide bond is cleaved by an enzyme the fluorescent endpoint of the free AMC can be detected.
Glycosynth offers a range of peptide/amide derivatives of AMC including L-Alanine-AMC.TFA (product code 33415) which is used for aminopeptidase activity, L-Leucine-AMC.HCl (product code 33340) for the determination of leucine aminopeptidase activity in waste water and L-Pyroglutamic acid 7-amido-4-methylcoumarin (product code 33158) which is used in an assay for the rapid identification of enterococci.
