Indoxyl Substrates

Indoxyl based substrates must be used under aerobic conditions to give the expected non-diffused coloured result. The non-diffuse end-point of these substrates makes them ideally suited for use in solid culture media.

Glycosynth offers a full range of substituted indoxyl substrates, offering a choice of coloured end-point. 5-bromo-4-chloro-3-indolyl or X-substrates (product code 70XXX) give a blue/green colour, 5-bromo-6-chloro-3-indolyl or mag substrates (product code 31XXX) give a purple colour, 6-chloro-3-indolyl or rose-substrates (product code 60XXX) give a pink colour, 5-bromo-3-indolyl substrates (product code 01XXX) give a deep blue end point, 3-indolyl substrates give a blue colour and 6-fluoro-3-indolyl or rouge substrates (product code 86XXX) offer a pale red colour.

5-bromo-4-chloro-3-indolyl alpha-D-galactopyranoside (product code 70039) is employed in the yeast two-hybrid screen for differentiating alpha-galactosidase-positive strains of yeast. For more information about the yeast two-hybrid screen see the detailed description on product 70039.

6-Chloro-3-indoxyl beta-D-galactopyranoside or rose-gal (product code 60045) is used to detect beta-galactosidase activity, giving red coloured colonies. Rose-gal can be used in conjunction with 5-Bromo-4-chloro-3-indolyl beta-D-glucuronide or X-glucuronide (our product code 70066 or 70065), a reagent to detect beta-glucuronidase, for the simultaneous detection of beta-glucuronidase and beta-galactosidase activity on the same diagnostic plate.
