Substituted MU substrates (CMU, TFMU, HDAMU)
Glycosynth offers several fluorogenic enzyme substrates based on the coumarin 4-methylumbelliferone. These substrates offer the advantage over 4-methylumbeliferyl counterparts that their lower pKa values mean they offer an increase in fluorescence intensities at physiological pH values.
The fluorescent substrate CMUG (6-chloro-4-methylumbelifery beta-D-glucuronide, product code 29064) shows superior performance over MUG (Product code 44064) when used for the detection of beta-glucuronidase producing E.coli in short-term assays. Compared with MUG, CMUG gives a much higher fluorescence with positive strains after 7h incubation.
6-Chloro-4-trifluoromethylumbeliferyl beta-D-galactopyranoside (product code 87045). Generates a relatively strong fluorescent signal after being hydrolysed by a target enzyme under neutral and acidic conditions.
6-Hexadecanoyl-4-methylumbellifery beta-D-galactopyranoside, (product code 62045) exhibits longer absorption/emission maxima (~385/500 nm) compared to 4-methylumbeliferyl beta-D-galactopyranoside (~365/450 nm). Offering a potential benefit when levels of endogenous fluorescence in the sample preclude the use of 4MU-substrates.
