Fluorogenic Substrates

Fluorogenic enzyme substrates are nonfluorescent compounds that consist of a fluorescent marker or fluorophore linked to an enzyme active group, typically a sugar, phosphate or ester that is recognized and acted upon by an enzyme to produce a detectable, soluble fluorescent end point. Fluorogenic substrates have a broad range of applications, including diagnostic microbiology, environmental monitoring, and in high-throughput screening to identify enzyme activity.

Fluorogenic Substrate Categories

The most widely employed fluorophores are based on hydroxy- and amino-substituted coumarins. Phosphate, ester and sugar derivatives of 7-hydroxy-4-methylcoumarin or 4-methylumbelliferyl (our product codes 44XXX) have been extensively used as fluorogenic enzyme substrates. Enzymatic cleavage results in the release of free 4-methylumbelliferone, which has a blue fluorescence upon excitation with UV light. The strongest fluorescence of 4-methylumbelliferone can be seen when the hydroxyl group is deprotonated at an alkaline pH. Maximum fluorescence intensity is observed with excitation at 350 to 370 nm and emission at 440 to 470 nm. Enzymatic cleavage of amide derivatives of 7-amino-4-methylcoumarin or AMC (our product code 33XXX) releases fluorescent AMC which can be seen with an excitation at 390nm and emission at 460nm.
